. Part of the book series (MIMB, volume 927) Abstract There are multiple clinical situations where cryopreservation of sperm is required including sperm banking prior to chemotherapy or radiation treatment for cancer, donor sperm for couples without a male partner, and various etiologies of male factor infertility. Bunge and Sherman presented the first pregnancy resulting from the thawing of cryopreserved sperm in 1953 (Bunge RJ, Nature 172:767–768, 1953). Starbucks create your own tumbler 16 oz template. Since that time, cryopreservation techniques have continued to improve. Here, we describe protocols for the cryopreservation and thawing of semen and testicular tissues using a simple liquid nitrogen vapor technique.

Although studies on sperm cryopreservation in marine mollusks started late in comparison with livestock and finfish, research has substantially increased for the major economical species farmed in the world, including oysters, mussels, scallops, pearl oysters, clams and abalone. In this paper, the latest developments in marine bivalves and gastropods sperm cryopreservation were reviewed and summarized for seven key steps: (1) sperm collection, (2) extender selection, (3) cryoprotectant agent selection, (4) sperm dilution/concentration before cryopreservation, (5) cooling rate, ( 6) thawing temperature, and (7) quality assessment. In general, the sperm quality is controlled by physiological and environmental parameters and further compromised by the cryopreservation processes. However, less attention has been paid to these issues in sperm cryopreservation in marine bivalves and gastropods.

Discussion about factors affecting sperm quality is, therefore, extended to include information from livestock and finfish species where they have been largely evaluated. Knowledge on these factors would not only improve our understanding of the key parameters influencing the success of sperm cryopreservation, but also provide information to develop strategies to improve the sperm quality in marine mollusks.
After your semen sample has been collected, we will place it on a warming block that’s 37°C so the sperm can turn into liquid. At this time, we will mix your semen sample in a 1:1 ratio with a freezing medium. This allows your sperm to survive the freezing and storage process. Frozen in Liquid Nitrogen Semen samples are slowly frozen in liquid nitrogen vapors. Once the samples have been frozen, they are placed in special containers where they are stored in the liquid nitrogen until they are needed.
Each ejaculate can yield between one to six vials. Each vial of a patient’s semen sample is labeled with the patient’s name, the date, and a unique number recorded on the side. This information will be used to catalog your semen sample in a storage facility. Test Thaw Procedure The survival rate of sperm after cryopreservation can vary widely. Depending on the amount of sperm you have put into a bank, we recommend that one of the cryovials be thawed so we can examine how successfully your semen can be stored and thawed.
1 Methods of sperm recovery Recovery of frozen spermatozoa by either IVF ( in vitro fertilization) or artificial insemination. Oocytes required for the IVF procedure are cultured after IVF until the two-cell stage has been reached. They are subsequently transferred into the oviduct of pseudopregnant foster females. Artificial insemination can be conducted as an alternative method to IVF.

Spermatozoa are directly injected into pseudopregnant females. This can be achieved by vaginal, intrauterine or direct transfer of sperm into the ampulla. In cases of poor fertilizing abilities, injection of a single sperm into the oocyte (ICSI, i.e. Intracytoplasmic sperm injection) can circumvent the problem. Another method called partial zona pellucida dissection (PZD) is used to improve IVF results in certain mouse strains, e.g.

The use of stallion frozen semen minimizes the spread of disease, eliminates geographic barriers, and preserves the genetic material of the animal for an unlimited time. Chrome speed booster 2013 dynamix rar file. Significant progress on the frozen thawed stallion semen process and consequently fertility has been achieved over the last decade.
These improvements not only increased fertility rates but also allowed cryopreservation of semen from “poor freezers.” This article reviews traditional steps and new strategies for stallion semen handling and processing that are performed to overcome the deleterious effects of semen preservation and consequently improve frozen semen quality and fertility.
